The OptiMyN project investigates the effects of Interferon-alpha therapy on myeloproliferative neoplasms (MPN), a type of blood cancer, aiming to optimize its therapeutic use. We apply a multi-scale mathematical modeling approach: differential equations for long-term patient treatment and stochastic models for short-term stem cell proliferation in vitro. Biological parameters are inferred using Bayesian, hierarchical, and approximate Bayesian computation (ABC) methods. The project, funded by ITMO Cancer – INSERM, brings together complementary expertise from Institut Curie (normal hematopoiesis), INSERM/Gustave Roussy (MPN specialists), and the biomathematics team (applied mathematics). Additional funding from Force Hémato supports the development of an online clinical decision support application integrating our methods.
This project aims at developing a unified mathematical model to understand and predict the progression from asymptomatic clonal hematopoiesis (CHIP) to myeloid blood cancers, particularly myeloproliferative neoplasms and secondary acute myeloid leukemia (AML). Using advanced whole-genome sequencing and statistical inference methods, our goal is to identify early biomarkers to detect individuals at risk and improve preventive strategies. The project involves collaboration with Isabelle Plo’s team at Gustave Roussy.
